Questions about NOD mouse diabetes.

چکیده
a still unknown triggering event such as the local inflammation brought by a putative virus or an antigen mimicry phenomenon. Alternatively hut not exclusively, it might involve a defect in protective immunoregulatory circuits. An intriguing possibility is that the onset of diabetes is secondary to the loss of the active tolerance that characterizes the 2-3-month-long period of clinically latent insulitis (prediabetic stage). If this is indeed the case, how does this tolerance operate (Thl/Th2 imbalance) ? What are the factors that control this loss of active tolerance ?

A susceptibility allele from a non-diabetes-prone mouse strain accelerates diabetes in NOD congenic mice.
The nonobese diabetic (NOD) mouse is genetically predisposed for the spontaneous development of type 1 diabetes. Linkage analyses have identified at least 19 susceptibility loci (Idd1-Idd19) that contribute to disease pathogenesis in which lymphocytes mediate the specific destruction of insulin-producing beta-cells. Interestingly, nondiabetic mouse strains have been shown to confer susceptibili...
متن کاملExpression of genetically determined diabetes and insulitis in the nonobese diabetic (NOD) mouse at the level of bone marrow-derived cells. Transfer of diabetes and insulitis to nondiabetic (NOD X B10) F1 mice with bone marrow cells from NOD mice
The development of autoimmune diabetes in the nonobese diabetic (NOD) mouse is controlled by at least three recessive loci, including one linked to the MHC. To determine whether any of these genetic loci exert their effects via the immune system, radiation bone marrow chimeras were constructed in which (NOD X B10)F1-irradiated recipients were reconstituted with NOD bone marrow cells. Unmanipula...
متن کاملExpression of somatostatin receptor subtypes 1-5 in pancreatic islets of normoglycaemic and diabetic NOD mice.
OBJECTIVE Somatostatin acts on five specific receptors (sst1-5) to elicit different biological functions. The non-obese diabetic (NOD) mouse is an experimental model of type 1 diabetes. The aim of this study was to investigate whether the islet expression of sst1-5 is affected during the development of diabetes in NOD mice, with insulitis accompanied by spontaneous hyperglycaemia. METHODS By ...

I n their article in this month's issue of Diabetes Care, Hummell et al. (1) tested the hypothesis that gluten is a driving antigen for type 1 diabetes–associated is-let autoimmunity. They convincingly demonstrated that elimination of dietary gluten for 12 months in humans positive for at least two type 1 diabetes–associated autoantibodies does not consistently alter the titers of these antibod...
متن کاملLocalization of Idd11 is not associated with thymus and nkt cell abnormalities in NOD mice.
Congenic mouse strains provide a unique resource for genetic dissection and biological characterization of chromosomal regions associated with diabetes progression in the nonobese diabetic (NOD) mouse. Idd11, a mouse diabetes susceptibility locus, was previously localized to a region on chromosome 4. Comparison of a panel of subcongenic NOD mouse strains with different intervals derived from th...
